Njoku was then ranked the 3rd best tight end available in the draft by Sports Illustrated and was ranked the 2nd best tight end by ESPN, NFL analyst Mike Mayock, and NFL analyst Bucky Brooks.
He was picked in the first round (29th overall) of the 2017 NFL Draft by the Cleveland Browns. Njoku was the third tight end to be picked in the draft.
2017 season
The Cleveland Browns signed him to a fully guaranteed, four-year, $9.52 million deal which included a signing bonus of 45.06 million on June 15, 2017. On September 10, he made his NFL debut in a 21-18 defeat by the Pittsburgh Steelers. Njoku had 2 receptions for 20 yards. He then had 3 receptions for 27 yards and his first NFL touchdown in the 24-10 defeat by the Baltimore Ravens in the Week 2 game. Njoku completed his rookie year with 32 catches for 386 yards and 4 touchdowns.
2018 season
Njoku joined the season as the Browns starting tight end in 2018. He played in all 16 games with 14 starts, posting 56 catches for 639 yards and 4 touchdowns.
2019 season
He was upended and landed on his head and left the game over the New York Jets because of a concussion in Week 2 of the 2019 season. However, it was announced that he suffered a broken wrist in the same play later in the week. Njoku was then placed on injured reserve on September 20, 2019. On November 20, 2019, he was designated for return from injured reserve and started practicing with the team again. Njoku was activated on December 7, 2019.
2020 season
The Browns exercised the 5th-year option on his deal on April 27, 2020. He was then placed on injured reserve with a knee injury on September 14, 2020, a day following posting 3 catches for 50 yards and a touchdown over the Baltimore Ravens. Njoku was later activated on October 10.
2021 season
Njoku caught 7 passes for 149 yards and a touchdown, with the score coming off a 71-yard catch and run in Week 5 over the Los Angeles Chargers.
2022 season
On March 7, 2022, the Browns placed the franchise tag on Njoku. He then signed a four-year, $56.75 million deal extension on May 27, 2022.
